Objective: To evaluate resting-state striato-cortical connectivity in PD and mild cognitive impairment due to Alzheimer’s disease (AD-MCI) and assess their relation to cognitive outcomes. We also aimed at directlz comparing groups with mild cognitive impairment due to different pathologies (PD-MCI and AD-MCI).
Background: Functional connectivity is abnormal in Parkinson’s disease (PD) and in mild cognitive impairment due to Alzheimer’s disease (AD-MCI).
Methods: Seed-based connectivity of the dorsal, middle, and ventral striatum was analyzed in 111 subjects using 3T fMRI. The correlation between connectivity at regions of between-group differences and clinical outcomes was also assessed.
Results: Patients showed lower striatal connectivity than HC. Connectivity between the middle (associative) striatum and precuneus negatively correlated with executive functions in PD and with memory performance in AD-MCI. PD-MCI compared to AD-MCI showed decreased connectivity of the dorsal (motor) striatum only.
Conclusions: Striatal connectivity was reduced in patients compared to HC. Similar compensatory mechanisms were employed to overcome various cognitive deficits in PD, PD-MCI and AD-MCI.
