Objective:
The present study has therefore been carried out identify neuroprotective role of quercetin in alleviating the cadmium induced dopaminergic dysfunctions via glial mediated deactivation of TNFα mediated CAMKIIα/ ERK signaling.
Background: Quercetin, a polyphenolic flavonoid, has been shown to play an important role in the treatment of neurodegenerative diseases including Parkinson’s Disease and Movement Disorders. Studies have shown that the microglial activation mediated toxicity is an important aspect wherein Quercetin have been shown to expert the anti inflammatory effects by attenuating the inflammatory mediators.
Method: The invivo studies were carried out using wistar rat with four different treatment groups. Protein expression of different signaling molecules were assessed using the western blotting, IHC and qRT-PCR techniques. The Behavioral studies to assess the dopaminergic dysfunctions were done using the Actimot, Y maze, Rota Rod apparatus, Grip Strengths. Neurodegenerative studies were carried out using the transmission electron microscopy.
Results: The rats treated with quercetin showed the significant downregulation in glial activation evident by IBA1 and GFAP Levels. Interestingly, A significant down regulations were observed at TNFα, CAMKIIα and P ERK levels on simultaneous exposure with quercetin. Quercetin also reduces the ROS production by decreasing the glial mediated toxicity in cadmium exposed rats.
Conclusion: The current study reveals that quercetin alleviates the cadmium induced dopaminergic alterations by hindering the glial activation through canonical TNFα/ CAMKIIα/ pERK signaling.
