Objective: This study aimed at investigating the mechanisms involved in the pathogenesis of Parkinson’s Disease in atrazine rat model of Parkinson’s Disease and the neuroprotective role of myricetin.
Background: Till date, atrazine is still being used for horticultural and agricultural purposes irrespective of the ban being placed on its usage. Parkinson’s Disease has been linked to the degeneration of dopaminergic neurons in the substantia nigra pars compacta. Myricetin, a natural compound and flavonoid is reputed to elicit quite a number of biological functions owing to its plethora of pharmacological properties.
Method: Adult male Wistar rats (250 g- 300 g) were divided into four groups of eight animals each and orally treated with atrazine, myricetin, and a combination of both at 50 and 20 mg/kg. b.w respectively for 45 days. Post treatment, markers of mitochondrial function, dopamine synthesis and transport, and Lewy body deposits were examined in the nigro-striatal system of the animals using immunological methods.
Results: Atrazine treatment significantly upregulated nigrostriatal expressions of PD genes, genes responsible for dopamine synthesis and transport, increased number of apoptotic neurons, and diminished the level of dopamine compared to the control. However, intervention with myricetin in the co-treatment group prevented these anomalies in the nigro-striatal system of the Wistar rats.
Conclusion: Result obtained from this study demonstrated that myricetin could be a potential therapeutic candidate in the treatment and management of Parkinson’s Disease associated with mitochondrial dysfunction, dopamine synthesis and transport, and apoptosis.
