Objective: We aim to explore the associations of IL-1β and IL-6 gene polymorphisms with the pathogenesis of PD and with clinical features of motor and non-motor symptoms.
Background: A wide variety of hypotheses have been put forward to explain the cause of neurodegeneration in PD, including inflammation. Both central and peripheral inflammatory immune responses have been documented in PD and they are thought to play a major role in the initiation and progression of PD. In recent years, it has been increasingly confirmed that gene polymorphism is one of the important factors affecting the susceptibility to various diseases , such as chronic obstructive pulmonary disease, lung cancer and PD.
Method: We have conducted a two-year observational cross-sectional retrospective study including patients diagnosed with idiopathic PD recruited from the neurological department of Charles Nicolle hospital and who underwent the genetic study of rs16944 at IL-1β-511, and rs1800795 at IL-6-174.
26 patients and 30 controls were included. Our patients were evaluated for motor impairment with the MDS-UPDRS-III and with Modified Hoehn and Yahr Scale. Overall non-motor symptoms were assessed for each patient, using NMS scale. To evaluate non motor symptoms we used: mini mental examination scale(MMSE), Frontal assessment battery(FAB), Beck Depression InventoryII (BDI-II), geriatric depression scale (GDS), Scales for Outcomes in Parkinson’s disease Autonomic Dysfunction (SCOPA-AUT), the gastrointestinal dysfunction scale for Parkinson’s disease (GIDS-PD) and RBD questionnaire.
Results: Our study revealed no significant difference in allele and genotype distributions between disease and control group for both IL-1β and Il-6 genes.
Mean UPDRS-III score and Mean H-Y scale were higher among IL-1β TT genotype comparing to CC and CT genotypes and it was higher in IL-6 CG genotype comparing to CC and TT genotypes but these findings weren’t statistically significant. No significant difference was detected between patients regarding motor phenotype.
We found no correlation between IL-1β and IL-6 gene polymorphisms and non motor symptoms.
Conclusion: The association of IL-6-174 and IL-1β-511 with PD is controversial, and contradictory results have been reported among previous studies and ours. Our study found no association of neither IL-6-174, nor IL-1β-511 with PD susceptibility in the Tunisian population.
