Objective: To study the neuroprotective efficacy of apocynin, a NADPH oxidase inhibitor in 1) alterations of post- stroke motor behaviour, 2) alterations of various neurotransmitter levels, 3) expressions of various inflammatory genes.
Background: The movement disorders following ischemia varies considerably from the day 1 of the onset of stroke to several months. From earlier studies, it was observed that chorea appears earlier (approximately 4 days post-stroke) while parkinsonism much later (approximately 117.5 days post-stroke). Various neurotransmitters play important role in post-stroke movement disorder. These motor deficits also well correlated to inflammatory cascades following ischemia.
Methods: To study these post stroke motor study, male wistar rats were subjected to ischemia by occlusion of both common carotid arteries for 15 minutes followed by reperfusion. Some animals were administered with apocynin (5 mg/kg bwt; i.p) 30 minutes prior to surgery and then daily up to 14 days. Different motor behavioural test were done at 5, 7 and 14 day; levels of dopamine, nor-epinephrine and serotonin was analysed by HPLC and the gene expression of various inflammatory cytokines by RT-PCR at day 14 following ischemic insult.
Results: Apocynin administration significantly improve the motor coordination and locomotor activity in ischemic animals. Also the alterations in levels of various neurotransmitters reversed to the control levels when compared to ischemic animals. The gene expression of various cytokines (IL-1α, IL-1β and TNF-α) were decreased significantly with apocynin treatment when compared to animals with ischemic insult.
Conclusions: The effect of Apocyanin seems to be mediated by normalizing of alterations in the brain monoaminergic and cholinergic systems, as well as inhibiting inflammatory cytokines. Which was seen by the improvement of motor activity and motor coordination.
