Objective: We performed a case-control study in Chinese population to evaluate the association of AD genome-wide association study (GWAS) -linked top hits with both PD and cognitive function in PD, investigating 13 single nucleotide polymorphisms (SNPs) in 9 genes (BIN1, CLU, ABCA7, CR1, PICALM, MS4A6A, CD33, MS4A4E and CD2AP).
Background: Alzheimer’s disease (AD), Parkinson’s disease (PD) and cognitive impairment in PD have overlapping clinical and pathological features, suggesting that there may be a genetic link for these diseases.
Methods: In total, 896 subjects consisted of 442 PD patients and 454 controls were genotyped. Out of all the PD patients, 75 was diagnosed as mild cognitive impairment (PD-MCI), 99 as dementia (PD-D). Logistic regression analysis was performed to calculate age- and sex- adjusted odds ratios.
Results: As a result, none of the AD – susceptibility loci showed statistically significant differences in allele, genotype and haplotype frequency distribution between the controls and PD cases, or between the controls and different PD subgroups stratified by cognitive function.
Conclusions: Our findings therefore imply that the 13 SNPs from AD GWAS may not play major roles in the genetic predisposition with PD and cognitive function in PD in a Chinese population.
