Objective: To investigate the features of beta oscillations of the subthalamic nucleus (STN) in the Parkinsonian minipig model.
Background: Deep brain stimulation (DBS) is an effective treatment for advanced Parkinson’s disease (PD) patients. In the past decade, the concept of closed-loop DBS was examined. The biomarker of closed-loop DBS in PD is primarily the abnormal features of local field potential (LFP) of the STN, that is, pathological beta oscillations. In large animal models, beta oscillations can be recorded in the STN of Parkinsonian non-human primates and minipigs. Minipigs have advantages in developing closed-loop DBS because they are easy to obtain and relatively inexpensive. However, the features of beta oscillations in the STN of PD minipig need further study.
Method: Three Lee-Sung breed minipigs were used in this study. The PD minipig model was induced by continuous injection of MPTP. PD symptoms were assessed by push test and hurdle test to evaluate postural instability and gait, respectively. The standard stereotaxic surgery was performed to implant DBS lead. After verifying the neuronal activities in the STN through microelectrode recording, a DBS lead was implanted into the STN. The other end of the lead was externalized in the back for recording. Three weeks after surgery, the PD minipigs were anesthetized with light Isoflurane (1.3-1.5%) to record the LFP of the STN, and spectral analysis was performed using fast Fourier transform.
Results: A cumulative dose of 10 mg/kg MPTP could induce PD symptoms in minipigs, assessed by push test and hurdle test, which are significantly different from those before injection. Beta oscillations were recorded in the STN of two of the three minipigs. Beta peaks were 21 and 25 Hz, respectively. Suppose a single threshold algorithm was considered for turning stimulation on and off. In this case, the power changes of beta peak ± 3 Hz in the frequency domain and the power changes of beta bursts in the time domain were compared, and the stimulation timing and duration were similar.
Conclusion: Clinical DBS lead can be used to record beta oscillations in the STN of the PD minipig model. The amplitude and spectral features of pathological LFP differential signals in PD minipigs are similar to those of PD patients.
