Objective: The objective of study is to find out the effect of isoquercitrin, a plant-derived chemical, against Parkinson-like symptoms induced by rotenone in zebrafish..
Background: Parkinson’s disease (PD) stands out as the most prevalent neurological disorder with a global impact. Currently, available medications only alleviate the clinical manifestations of PD, offering no substantial impact on slowing down the disease’s progression. Rotenone is a neurotoxic pesticide that causes PD by specifically harming dopaminergic neurons. Isoquercitrin, a rare flavanol glycoside and a principal glycosidic form of the natural flavanol quercetin, exhibits diverse biological functions.
Method: Zebrafish were exposed to daily doses of rotenone (5 μg/L) and isoquercitrin (25 & 50 mg/kg) for 7 days. On day 8, zebrafish were sacrificed for the evaluation of oxidative stress parameters (lipid peroxidation, nitrite and reduced glutathione, glutathione peroxidase, superoxide dismutase, catalase), neuroinflammatory cytokines, mitochondrial complexes I and IV, neurotransmitters (brain catecholamines, gamma-aminobutyric acid, glutamate) and histopathological examination.
Results: Rotenone induced significant motor deficits, as evidenced by behavioural changes, increased oxidative stress, elevated levels of neuroinflammatory cytokines, altered neurotransmitter levels, dysfunctional mitochondrial complexes, and histopathological abnormalities. Isoquercitrin (25, 50 mg/kg) demonstrated a dose-dependent improvement against rotenone-induced behavioural abnormalities. It reduced oxidative stress and reversed aberrant neurotransmitter changes in the striatum, inflammatory responses and dopaminergic neuron degeneration in striatum.
Conclusion: These results underscore the considerable anti-inflammatory and antioxidant properties of isoquercitrin, highlighting its robust neuroprotective effects against rotenone-induced neurotoxicity.
