Objective: Parkinson’s disease can cause neuropsychiatric disturbances which can range from mild to severe. This includes disorders of speech, cognition, mood, behaviour, and thought. In addition to cognitive and motor symptoms, PD can impair other body functions. Sleep problems are a feature of the disease and can be worsened by medications. Sleep and wakefulness are behavioral and physiological activities. It is a modified form of the basic rest activity cycle.
Background: The present study has been initiated to find out the effect of Norepinepherine and Serotonin and the blockers Prazosin and Propranolol in different permutations and combinations on Sodium Potassium ATPase activity along with insilico drug repurposing to find their efficacy in treating PD.
Method: Male wistar rats weighing 250-280 gms, obtained from central animal house facility of Jawaharlal Nehru University were used. The animals were kept in semitransparent polypropylene cages and were maintained under 12:12 dark: light cycle. Room temperature was maintained at 25-28°C. Food and water were supplied ad libitum. Brain from the male wistar rats weighing 250-280 gm was extracted and subjected to homogenization, synaptosome was prepared and Na-K ATPase activity was estimated under the influence of NE (100μM), 5HT(μM), Prazosin (5mM) and Propranolol(5mM) in different combinations.
Results: Both NE and 5HT increase the Na-K ATPase activity individually and also synergistically when used in combination but in presence of receptor antagonists a decrease is observed. Moreover Prazosin and Propranolol also decrease the basal values of Na-K ATPase activity.
Conclusion: We included Insilico studies for the first time using Schrodinger’s Software and introducing the concept of drug repurposing which provided us a new path in exploring the therapeutic possibilities for some of the Parkinson’s and other movement disorders.
